No it wont.

Think you are talking about the "spine, or backbone" of the rod.
Most rods will have 1, some have 2. To find it, put the rod but on the ground, and hold the tip up to about 45 deg.
Press down about 1/3 from the tip with 1 finger, and put a bend in the rod. The rod will twist untill it settles in one position.
Thats the backbone, and the guides should be bound on that position.
Heaps of rod builders have got it back to front and put the guides in the opposite position, and theres no worries doing that.
On really heavy rods, like 130lb and 80lb Game fishing rods, if the guides are not built on the bone, one way or the other, then the rod will twist, eventually wrecking the fancy binding on the guides, but it wont break.

Some really good rods dont have a back bone. a good example is the top of the line St Croix,s like legend elite.
This due to the manufacturing process of thr blank. Back bones are formed where each wrap of graphite, or what ever material is used on the rod meets the other half of the material, or over laps. It wont cause the rod to break, maybe twist a little under load. I doubt if anyone could notice it on a baitcaster or light spin rod. Have look at Bushy,s spiral wrap rods.
